To be able to observe individual individual repeats in a protein using standard NMR spectroscopy methods, it is crucial that the protein is sufficiently stable to allow for multiple measurements per residue, and that the repeat unit gives a distinctive spectrum. In this study we provide a method for determining whether a given repeat yields a distinct and stable NMR spectrum. Two repeats were selected based on our previous experience, representing a terminally truncated and a full-length protein construct. These proteins were studied using standard and anisotropic mixing methods. The relative intensities of all the multiplets in the spectra of each of the proteins were then measured, and the repeat unit assignments were determined. The repeat unit conformations were then modified and analysed to determine the effect of the perturbation. While the repeat unit of the terminally truncated construct and the repeat unit of the full-length construct both yielded distinct spectra, a significant change in the spectrum was observed with the addition of the second repeat. The repeat unit of the full-length construct had a conformation with a high degree of dispersion, while the repeat unit of the truncated construct had a conformation with fewer and more condensed residue interactions. It is concluded that there is a correlation between the repeat unit conformation and the stability of the domain, which can be obtained from the NMR data.Electric-field modulated quantum-dot infrared photodetector. We show that the presence of the gate electrode as a nanoscale load electrode shifts the interdot state energy levels of nanoscale quantum-dot mid-infrared photodetectors (QDMIP) due to the capacitive effect of the gate electrode. There are two mid-infrared photodetectors, which are biased in reverse polarity when one terminal of the gate electrode is grounded and in the same polarity when the other terminal of the gate electrode is grounded. The photocurrent is significantly modulated by the gate E-field under reverse bias. The photocurrent change in the QDMIP increases with the gate E-field.
